# TUTORIAL: Height control of PEG hydrogels with HyPE and PRIMO

HyPE is a photopolymerization solution enabling the control of the height of hydrogel structures.

This tutorial describes the full 4-Arm-PEG-Acrylate photopolymerization protocol using HyPE 400, the PRIMO2 photopatterning system and the LEONARDO5 software.
It allows to structure photosensitive hydrogels in 3D for further cell culture:

– Hydrogel solution preparation
– PRIMO 2 objective calibration
– Use of the Hydrogel structuration module of the new LEONARDO 5 software version
– Optional: Height characterization using fluorescent beads
